    HLA-E, a non-classical major histocompatibility complex (MHC) molecule, has various applications in different fields. In the context of gestational diabetes mellitus (GDM), HLA-E is identified as one of the differentially expressed genes (DEGs) in placenta tissues of GDM patients. Its down-regulation suggests a potential role in GDM prevention and management. In Alzheimer's disease (AD), HLA-E is involved in the abnormal signal transduction of major histocompatibility complex class I (MHC-I) and is associated with increased content of GZMK+ CD8+ T cells in the peripheral blood. These GZMK+ CD8+ T cells, along with their interactions with other cell types, may serve as potential blood biomarkers of AD. In nodular lymphocyte predominant Hodgkin lymphoma (NLPHL) and T-cell/histiocyte-rich large B-cell lymphoma (THRLBCL), the immune cell phenotypes accompanying different histological patterns involve expansions and depletions of specific immune cell subsets, including HLA-E-expressing cells. These findings provide insights into the immunoregulatory mechanisms and potential biomarkers of high-risk disease in NLPHL and THRLBCL. In the context of influenza virus infection, a cryptic MHC-E epitope derived from the influenza matrix gene segment elicits a potent cytolytic T cell response. This unconventional epitope, presented by MHC-E, highlights the importance of noncanonical translation products in the T cell repertoire and suggests therapeutic potential. Lastly, in human cytomegalovirus (HCMV) surveillance, HLA-E-restricted HCMV peptides are recognized by adaptive NK cells and induce memory responses. These unconventional peptides elicit aNK cell memory responses restricted to HCMV and provide insights into HLA-E-mediated signaling networks that mediate aNK cell recall responses. Overall, HLA-E has diverse applications in GDM, AD, lymphoma, viral infections, and immune cell memory responses.
