Many existing drugs acting on MPs have significant drawbacks, such as lack of specificity due to similar ligand-binding pockets in related MPs, or unwanted effects due to their involvement in a variety of signaling pathways that can have a significant impact on overall cell equilibrium conditions, such as ion channels and transporters. These can manifest as negative safety and side-effect profiles, highlighting the need for treatments that target specific MP conformations or signaling pathways, allowing for increased drug affinity, specificity, and potency, among other benefits.
