    Applications
The human CSF1R gene encodes the cell-surface protein known as colony stimulating factor 1 receptor (CSF1R), sometimes referred to as CD115 and macrophage colony-stimulating factor receptor (M-CSFR). Two ligands, colony stimulating factor 1 (CSF-1) and interleukin-34 (IL-34), can activate the CSF1R receptor. Myeloid cells have high expression levels of CSF1R, and both in vivo and in vitro myeloid cell types cannot survive, proliferate, or differentiate without CSF1R signaling. Treatments for cancer, neurodegeneration, and inflammatory bone diseases all target CSF1R signaling, which is implicated in a variety of diseases. The customized CSF1R stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

Fig.1 CSF-1R localizes in the nucleus of SKBR3 breast cancer cells.

Localization of CSF-1R in SKBR3 cells using cell fractionation. As previously mentioned, 50 nM siCONTROL non-targeting pool or 50 nM SMART-pool small interfering RNA (siRNA) targeting CSF-1R mRNA were used to achieve gene silence.

    Research Highlights

This study examines the processes relating CSF1R mutations to pleiotropic effects on postnatal growth and development, as well as the evidence that CSF1R is exclusively expressed in mononuclear phagocytes.

A patient with a frameshift mutation exhibits haploinsufficiency of the colony-stimulating factor-1 receptor (CSF1R), which makes it easier to develop a Csf1r haploinsufficient animal model. The pathologic, radiologic, and clinical characteristics that these animals exhibit are similar to those of CSF1R-mutant human patients.
