Migration and invasion capabilities of colon cancer cell lines were assessed via the transwell assay (as depicted in the right panels). The expression levels of STK4 were measured under conditions of STK4 knockdown or overexpression, as shown in the left panels. The data were presented as mean ± SD, based on three independent experiments. In H3347 cells, a notable increase in migrated cells was observed in the STK4-shRNA group (81 ± 18) compared to the Scr group (24 ± 12, P = 0.00006). Similarly, in the STK4-shRNA group, invaded cells were significantly higher (39 ± 13) compared to the Scr group (12 ± 4, P = 0.0062). In DLD-1 cells, the GFP-STK4 group exhibited reduced migration (18 ± 4) compared to the GFP group (34 ± 5, P = 0.037). Additionally, invaded cells in the GFP-STK4 group (33 ± 8) were significantly lower compared to the GFP group (60 ± 11, P = 0.041). The scale bar denotes 200 μm. The Mammalian STE20-like kinase 1 (MST1/STK4/KRS2) is a serine/threonine kinase that is highly conserved and a key member of the Hippo signaling pathway. Previous research has shown its significance in different types of tumors, yet its specific role in clear cell renal cell carcinoma (ccRCC) has not been thoroughly investigated. In this study, the function of STK4 in ccRCC will be systematically and comprehensively examined through various experiments and analyses.

Serine/threonine kinase 4 deficiency (STK4 or MST1, OMIM:614868) is an autosomal recessive (AR) combined immunodeficiency that can manifest with epidermodysplasia verruciformis-like lesions (EVLL) on the skin. In this study, a 17-year-old male patient, born from consanguineous parents, is presented. He has a history of recurrent respiratory infections, verruciform skin plaques, poikiloderma, chronic benign lymphoproliferation, and Sjögren syndrome. He is suspected to have interstitial lymphocytic pneumonia.
