    Applications
P2RY8 is a transmembrane receptor that binds to and is activated most selectively by adenosine and uridine nucleotides. Adenosine and uridine nucleotides stimulate P2RY8 activity, which in turn stimulates the intracellular signaling pathway for adenylyl cyclase. P2RY8 can activate phospholipase C and the extracellular signal-regulated kinases signaling pathway after binding to nucleotides. There are also several studies that demonstrate the role of purinergic receptor signaling in a number of biological processes, including cell migration, proliferation, differentiation, and apoptosis. P2RY8 may play a role in leukemogenesis as patients with high levels of P2RY8 expression have a higher chance of developing leukemogenesis than individuals with low levels of P2RY8 expression. The customized P2RY8 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Research Highlights

A major source of B cell malignancies, germinal centers are significant locations for antibody diversification and affinity development. Uncertainty surrounds the cues that encourage this restriction. A Gα13-coupled receptor called P2RY8 controls the expansion of B cells in lymphoid organs and mediates their restriction of migration.

P2RY8 overexpression in cells and animals inhibited the growth of plasma cells and strengthened the negative selection of developing B cells with DNA reactivity. These findings reveal a function for P2RY8 in the development of lupus and immunological tolerance.
