    Applications
The MAP4K5 gene in humans codes for the enzyme mitogen-activated protein kinase kinase kinase 5. The protein kinase belonging to the serine/threonine family that is encoded by this gene bears a striking resemblance to yeast SPS1/STE20 kinase. Near the start of the MAP kinase signal cascades, which are crucial for the yeast pheromone response, is where yeast SPS1/STE20 functions. In mammalian cells, it was demonstrated that this kinase activated Jun kinase, suggesting a function in the stress response. For this gene, two alternatively spliced transcript variants that encode the same protein have been identified. The customized MAP4K5 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Published Data

Fig.1 Immunoblotting for MAP4K5 and E-cadherin expression in HPDE cells.

Immunoblotting immortalized human pancreatic ductal cells, pancreatic cancer cell lines, and HPDE cells for the expression of MAP4K5 and E-cadherin. The MAP4K5 cDNA construct-transfected 293T cells were preserved as a positive control for immunoblotting.

    Research Highlights

In most PDACs, MAP4K5 expression is either absent or reduced. A significant involvement for MAP4K5 in the epithelial-to-mesenchymal transition, chemotherapy resistance, and tumor progression in pancreatic cancer may be suggested by the high correlations shown between low MAP4K5 expression and loss of E-cadherin, lower CES2 expression, and poor overall survival.

The complicated signaling cascade known as the STE20 kinase family controls cytoskeletal organization and affects the stress response. MAP4K5 and TAOK1 are two examples of the kinase mediators involved in this signaling cascade.
