    The 56 kDa protein Lck is present in lymphocytes, which are specific immune system cells. As a member of the Src kinase family (SFK), Lck plays a crucial role in naive T cells as well as effector T cells in activating T-cell receptor signaling. Compared to CD4 T cells, the activation or maintenance of memory CD8 T cells is less dependent on the Lck. Additionally, different memory T cell subsets have different roles for the lck. It appears that in mice, among the population of effector memory T cells, over 50% of lck exists in a constitutively active conformation, while less than 20% of lck exists in an active form. The customized LCK stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Western blotting was used to evaluate the total protein expression and flow cytometry was used to analyze FLT3 surface expression. It is an appropriate model to investigate the effect of LCK on FLT3-ITD mediated biological events because FLT3 surface and total expression seems to be the same for both LCK and empty vector expressing cells.
