    Applications
The LATS2 gene in humans encodes the enzyme large tumor suppressor kinase 2 (LATS2). This gene participates in the Hippo signaling pathway, where it inactivates the effector proteins, YAP and WWTR1 (TAZ). It produces a serine/threonine protein kinase that is a member of the LATS tumor suppressor family. In both early and late metaphases as well as interphase, the protein localizes to centrosomes. It interacts with the centrosomal proteins ajuba and aurora-A and is necessary for spindle formation and gamma-tubulin accumulation at the beginning of mitosis. It may work in a positive feedback loop with p53 in response to cytoskeleton damage. It also interacts with a negative regulator of p53. It can also act as a corepressor of the expression of genes that respond to androgen. The customized LATS2 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Published Data

Fig.1 LATS2 alterations in MPM cells in culture.

LATS2 mRNA and protein were expressed by MPM without mutation. Only those MPM exhibiting significant exon deletions showed complete reduction of mRNA expression. Only the MPM in culture with missense mutation (MPM_04) displayed LATS2 protein expression out of the seven LATS2 mutants.

    Research Highlights

According to available data, the MOB1 scaffold protein and the hippocampal kinases MST1/2 encourage the development of active MOB1/LATS complexes, which phosphorylate and inhibit YAP1.

According to this study, LATS2 O-GlcNAcylation plays a crucial part in the deregulation of the Hippo pathway in cancer cells, which is directly linked to the formation of tumors.
