    Applications
It was discovered and isolated a novel potassium channel gene in humans. A protein with 456 amino acids is encoded by the maximum open reading frame. The murine voltage-gated potassium channel protein Kv1.7 (Kcna7), which is crucial for the repolarization of cell membranes, shares 91% of its amino acids with the expected product. The human gene has been identified as the ortholog of the mouse Kcna7 and given the name Kv1.7 (KCNA7) due to their high resemblance. A structural prediction revealed six membrane-spanning domains and a pore area typical of potassium channels. The gene is expressed preferentially in skeletal muscle, the heart, and the kidney, according to Northern expression analysis. However, other tissues, such as the liver, express it at a lower level. There was only one mRNA isoform found, and its size was approximately 4.5 kb. The customized KCNA7 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Published Data

    Fig.1 RT-PCR analysis of KCNA7.

With the use of KCNA7-specific primers, RT-PCR analyses were carried out to verify the existence of KCNA7 mRNA in the adult human heart. These findings demonstrate that the mature human heart expresses KCNA7.

    Research Highlights

Methylation levels of nine candidate genes and LINE-1 repeats were quantified by bisulfite pyrosequencing in sperm DNA of 162 donors and 191 cord blood samples of resulting children.
