    Serotonin's G-protein coupled receptor, HTR1E (5-HT1E), performs this function. Additionally, it can serve as a receptor for several alkaloids and psychotropic drugs. When 5-HT1E interacts with its ligand 5-hydroxytryptamine, a conformational shift occurs that initiates signaling via guanine nucleotide-binding proteins and controls the activity of downstream effectors such adenylate cyclase. Adenylate cyclase activity is inhibited by 5-HT1E activation. It is possible to use 5-HT1E as a drug target for the treatment of neuropsychiatric disorders characterized by memory deficits or as a target for the treatment of temporal lobe epilepsy due to the stimulation of 5-HT1E and subsequent inhibition of adenylate cyclase activity in the dentate gyrus (DG). This finding suggests that 5-HT1E may mediate regulation of hippocampal activity by 5-HT. The customized HTR1E stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Keep in mind that engineered cells are known to reproduce more slowly than their unmodified mother cells. Additionally, confirm that you are using the appropriate media reagents for your cell line. If so, the following are some additional frequent causes:
    Adding antibiotics to a thawed culture prematurely.
    Spinning down the thawed cells to remove DMSO.
    Over-splitting.
    Clumping.

    Examine the condition of your cells. Verify that all culture or plating methods were followed and that the cells arrived frozen.
    The high protein concentration of crude lysates may be hindering the experiment if you're utilizing them.
