    Applications
The human CDK8 gene encodes the enzyme cell division protein kinase 8. This gene codes for a protein that belongs to the family of cyclin-dependent protein kinases, or CDKs. Through many processes, CDK8 and cyclin C regulate transcription by forming an association with the mediator complex. Many transcription factors are bound by CDK8 and/or phosphorylated, which can either activate or inhibit the action of the transcription factors. SREBP, STAT1 S727, and the Notch intracellular domain are all phosphorylated by CDK8. Additionally, through affecting the turnover of subunits in the mediator complex tail module, CDK8 prevents transcriptional activation. Furthermore, RNA polymerase II's interaction to the mediator complex is influenced by CDK8. The customized CDK8 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Published Data

Fig.1 Western blot analysis of colon cancer cell lines for CDK8 and β-catenin.

Every cell line exhibited strong expressions of β-catenin and CDK8. Each cell line showed an increase in β-catenin levels but a decrease in CDK8 levels following a 24-hour serum deprivation.

    Research Highlights

These findings imply that, in line with CDK8's function in chemotherapy-induced tumor-supporting paracrine activities, the expression of CDK8 and its interacting genes has a significant influence on the response to adjuvant therapy against breast cancer.

By joining the mediator complex or phosphorylating transcription factors (TF), cyclin-dependent kinases 8 (CDK8) controls the transcriptional process. Numerous malignancies have been reported to overexpress CDK8.
