    Humans have an enzyme called cyclin-dependent kinase 7, or cell division protein kinase 7. The CDK7 gene encodes this enzyme. This gene codes for a protein that belongs to the family of cyclin-dependent protein kinases, or CDKs. Members of the CDK family, who are known to be significant regulators of cell cycle progression, are very similar to the gene products of Saccharomyces cerevisiae cdc28 and Schizosaccharomyces pombe cdc2. Together with cyclin H and MAT1, this protein forms a trimeric complex that activates Cdk (CAK). It is a crucial part of the transcription factor TFIIH, which is involved in DNA repair and transcription initiation. It is believed that this protein acts as a direct conduit between the cell cycle and transcription control. The customized CDK7 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    Phosphorylation of RNAPII's CTD contributes to transcription initiation and RNAPII procession, and CDK7 kinase activity is implicated in this process. Similar effects of THZ1 were seen in H1975 and H1975/OR cells on RNAPII-CTD activity. At the lowest concentration of THZ1, the H1975/WR cells, however, showed suppression of RNAPII-CTD phosphorylation at Ser2, Ser5, and Ser7. Furthermore, EGFR or Akt activity was not inhibited by THZ1 therapy, whereas Erk activity exhibited a dose-dependent increase.

    In addition to being an effector CDK that phosphorylates Pol II and other targets within the transcriptional machinery, Cdk7 is also a CDK-activating kinase (CAK) for at least one other crucial CDK implicated in transcription. Cdk7 is linked to the transcription initiation factor TFIIH.
