Fig.1 Effects of CDK7 inhibitors on cells with acquired resistance to WZ4002 or osimertinib.
Phosphorylation of RNAPII's CTD contributes to transcription initiation and RNAPII procession, and CDK7 kinase activity is implicated in this process. Similar effects of THZ1 were seen in H1975 and H1975/OR cells on RNAPII-CTD activity. At the lowest concentration of THZ1, the H1975/WR cells, however, showed suppression of RNAPII-CTD phosphorylation at Ser2, Ser5, and Ser7. Furthermore, EGFR or Akt activity was not inhibited by THZ1 therapy, whereas Erk activity exhibited a dose-dependent increase.

Research Highlights
Cyclin H, RING-finger protein MAT1, and cyclin-dependent kinase 7 (CDK7) combine to create the heterotrimeric CDK-activating kinase (CAK) complex, which is essential for transcription and cell-cycle regulation.

In addition to being an effector CDK that phosphorylates Pol II and other targets within the transcriptional machinery, Cdk7 is also a CDK-activating kinase (CAK) for at least one other crucial CDK implicated in transcription. Cdk7 is linked to the transcription initiation factor TFIIH.
