Background of 5-Hydroxytryptamine Receptors
5-hydroxytryptamine (5-HT) is a monoamine neurotransmitter and local hormone generated from L-tryptophan in serotonergic neurons and enterochromaffin cells along the gastrointestinal tract. It is the endogenous ligands of 5-hydroxytryptamine receptors. The family of 5-HT receptors is a group of G protein-coupled receptors. 5-HT receptors are classified into 6 groups, including 5-HT1, 5-HT2, 5-HT4, 5-HT5, 5-HT6, and 5-HT7 receptors.

Distributions and Functions of 5-Hydroxytryptamine Receptors
5-HT1 receptors are distributed throughout the central nervous system. 5-HT2 receptors are expressed in the brain, heart, and serotonin neurons, which play an important role in psychotropic pharmacology for antipsychotics, increased appetite and metabolic syndrome, and migraine. Agonists of 5-HT4 receptors are effective drugs for gastroesophageal reflux, constipation, and irritable bowel syndrome. Agonists of 5-HT6 and 5-HT7 receptors are non-selective drugs used to treat schizophrenia.
Subtypes and Mechanisms of 5-Hydroxytryptamine Receptors
5-HT receptors are a group of G protein-coupled receptors classified into 6 groups, including 5-HT1, 5-HT2, 5-HT4, 5-HT5, 5-HT6, and 5-HT7 receptors. There are several subtypes in each group. The group of 5-HT1 receptors contains five subtypes of receptors, coupling to Gi/Go proteins, which inhibit adenylyl cyclase. The group of 5-HT2 receptors contains three subtypes of receptors, coupling to Gq/G11 proteins, which stimulate phospholipase C. 5-HT4, 5-HT6, and 5-HT7 receptors couple Gs proteins. 5-HT5 receptors couple Gi/Go proteins and Gq/G11 proteins.

Abstract
|
In Schistosoma mansoni, the myoexcitatory action of 5-hydroxytryptamine (5-HT) that regulates muscle contraction in parasitic flatworms is through the activation of a serotonergic GPCR (Sm.5HTRL). Therefore, the study aimed to discover anthelmintic drugs targeting the Sm.5HTRL to control muscle contraction in parasitic flatworms. They used a cAMP biosensor assay to examine the effects of several aporphine alkaloids on the signaling activity of a heterologously expressed Sm.5HTRL construct. They found that the natural product nuciferine may act as an effective inhibitor of adult worm motility.

They have found four aporphine alkaloid derivatives with similar structures, nuciferine, D-glaucine, boldine, and bulbocapnine acting as 5-HT receptor antagonists. The four compounds were demonstrated to block Sm.5HTRL evoked cAMP generation. The potency of GPCR blockade was closely related to the ability of each drug to inhibit the contractility of schistosomula larvae. The results showed that the potency of GPCR blockade correlated well with the inhibition of schistosomule contractility. It was demonstrated that nuciferine was also effective at inhibiting both basal and 5-HT evoked motility of adult schistosomes, which suggested the effectiveness of Sm.5HTRL antagonists as hypomotility-evoking drugs across different parasite life cycle stages.
