    TAOK3, a STE20-type kinase, has been studied in various contexts. In the context of non-alcoholic fatty liver disease (NAFLD) and obesity, TAOK3 deficiency in mice did not affect the development of NAFLD or systemic insulin resistance. However, in vitro studies showed that TAOK3 knockdown suppressed ectopic lipid accumulation and metabolic stress in hepatocytes. In esophageal squamous cell carcinoma (ESCC), TAOK3 was found to promote cancer progression and cisplatin resistance by augmenting autophagy. In the context of age-associated inflammation, TAOK3 was found to limit inflammation by negatively modulating macrophage differentiation and their production of TNFα. Additionally, TAOK3 was found to regulate type 2 immunity and metabolism in obesity by inhibiting adipose tissue Tregs and regulating immunometabolism. Finally, a transcriptome-wide association study implicated TAOK3 as a candidate gene for depression, with its expression altered in specific cell subtypes in the dorsolateral prefrontal cortex. These findings highlight the diverse roles of TAOK3 in various biological processes and diseases.
