    The MAP3K4 gene in humans encodes the enzyme mitogen-activated protein kinase kinase 4. A conserved cascade of three protein kinases forms the basic core of every mitogen-activated protein kinase (MAPK) pathway: an active MAPK kinase kinase (MAPKKK) phosphorylates and activates a particular MAPK kinase (MAPKK), which in turn activates a particular MAPK. Environmental pressures activate JNK MAPKs and CSBP2 (p38α) MAPKs, whereas mitogenic stimulation activates ERK MAPKs. The MAPKKK, or MEKK4 protein, also known as MTK1, is encoded by this gene. At the C terminus of this protein lies a protein kinase catalytic domain. A regulatory domain can be present in the N-terminal nonkinase domain. In mammalian cells, MEKK4 expression triggered the JNK MAPK and CSBP2 (p38α) pathways, but not the ERK pathway. The customized MAP3K4 stable cell line can be used in antibody discovery and development, potential drug candidate screening and signaling pathway researches.

    After lysates were exposed to either anti-MAP3K4 or anti-MAP3K4 antibody, Protein A-Sepharose was used to precipitate the mixture. Proteins that were immunoprecipitated were separated using SDS-PAGE. Using the designated antibodies, proteins were immunoblotted after being transferred to nitrocellulose membrane.

    Bipotential gonads must commit to either an ovarian or a testicular fate in order to determine sex. In XY mice, gonadal sex reversal is caused by gene deletion of the kinase Map3k4, and the sex reversal phenotype is restored by transgenic re-expression of Map3k4.
