    IL2RA, also known as interleukin 2 receptor subunit alpha, has been studied in various medical contexts. In bipolar disorder (BD), IL2RA rs2104286 and IL2 rs2069762 polymorphisms have been found to be associated with the disorder and its clinical features. The study suggests that immuno-inflammatory pathways in the brain and environment contribute to the development of BD. In multiple myeloma (MM), IL2RA rs2104286, IL-2 rs2069762, and rs2069763 polymorphisms have been investigated for their effects on disease susceptibility, progression-free survival (PFS), and overall survival (OS). The study found that the IL2 rs2069762 TG genotype was associated with shorter PFS and OS, while the IL2 rs2069763 GG genotype was associated with shorter PFS. In kidney transplant recipients, the use of lymphocyte-depleting antibodies, such as anti-interleukin-2-receptor antibodies (IL2RA), has been examined in specific populations. The study found that compared to IL2RA, anti-thymocyte globulin (ATG) reduced the risk of acute rejection in standard and high-risk KTR, but did not decrease the risk of graft loss. The optimal induction therapy for kidney transplant recipients is still uncertain and requires further study. In oral cavity squamous cell carcinomas (OSCC), DNA methylation differences within INS, PTPN22, and IL2RA promoters have been observed in lymphocyte subsets. These differences in DNA methylation may contribute to an immunosuppressive tumor microenvironment in OSCC. The study also identified immune-associated long non-coding RNAs (lncRNAs) that may be involved in OSCC. In children with type 1 diabetes (T1D), the effect of T1D-susceptibility associated single nucleotide polymorphism (SNP) markers in INS, PTPN22, and IL2RA on CpG site methylation of their promoters in lymphocyte subsets was investigated. The study found associations between rs689 in INS and methylation at four CpG sites, as well as an association between rs12722495 in IL2RA and methylation at two CpG sites in B cells. These findings suggest that DNA methylation may mediate the genetic effects of these SNPs on T1D pathogenesis. Overall, IL2RA has been studied in the context of bipolar disorder, multiple myeloma, kidney transplantation, oral cavity squamous cell carcinomas, and type 1 diabetes, providing insights into its potential role in these conditions.

    Immunoblot analyses were conducted to assess the abundance of key regulators involved in cell proliferation/survival, cell-cycle progression, and apoptosis in UCSD/AML1 and HL60 derivative cell lines. These experiments were carried out three times, yielding comparable results. Activating phosphorylations on Thr202 and Tyr204 were observed in ERK, while Ser473 exhibited activating phosphorylation in AKT, and Tyr694 in STAT5.

The study included 200 BD patients and 200 healthy controls. The findings suggest that immuno-inflammatory pathways in the brain and environment may play a role in the etiology of this chronic affective disorder.

    Interleukin-2 (IL-2) is a cytokine secreted by T helper type 1 cells and released after induction of T helper cells with major histocompatibility complexes or antigens presented by antigen presenting cells. Its activity and gene polymorphisms have been extensively studied in both solid and hematological malignancies. This study aimed to investigate the effects of IL-2RA rs2104286, IL-2 rs2069762 and rs2069763 polymorphisms on multiple myeloma (MM) susceptibility, progression-free survival (PFS) and overall survival (OS). A total of 300 MM patients and 170 healthy individuals were included, and the genotypes of the polymorphisms were compared to reveal their effects on disease susceptibility and survival. The results showed that patients with the TG genotype of IL-2 rs2069762 had a significantly shorter PFS and OS compared to others, while those with the GG genotype of IL-2 rs2069763 had a shorter PFS. These findings could have implications for IL-2 based therapies and MM epigenetics.
