Pseudovirus Products

Pseudoviruses are viral particles covered with a heterologous envelope protein that facilitates entry of the pseudoviruses just as effectively as the entry of real viruses with great pathogenicity and infectivity. Pseudoviruses are unable to replicate on their own and can only infect host cells for one cycle as a result of the loss of the envelope protein gene and the lack of pathogenic genes. Pseudoviruses also provide the desired qualities of high safety, robust operability, and ease of use for high-throughput detection. Our HPV Pseudovirus

HPV virions are made up of icosahedral capsids that carry dsDNA and are not enclosed. Circular, 8 kb in size, and replicated inside host cell nuclei are the HPV viral genomes. The most effective method of producing PsV is the transfection of reporter (fLuc, SEA or GFP)- and L1- and L2-expression plasmids into 293T or 293FT cells. This method enables the efficient intracellular synthesis of papillomavirus gene transfer vectors. When a 5.9-kb plasmid is encapsidated, PsV self-assemble within the transfected producer cells, allowing for the creation of many billion transducing units per milliliter; whereas, an 8-kb plasmid results in 10-fold fewer transducing units per milliliter.

Advantages of Pseudovirus

Research on some viruses should be conducted in a Biosafety Level (BSL)-3 laboratory due to their high pathogenicity and infectivity, which restricts the creation of effective antiviral defenses. Pseudoviruses are thought to be safe and effective virological tools, because they include the essential components of virus infection and cell entry but lack nucleic acid and lose the ability to self-replicate. PsV are structurally similar by cryoelectron microscopy and have similar immunoreactive profiles against conformation-dependent neutralizing antibodies, suggesting that some, if not all, surface conformational epitopes are retained in PsV. This is in addition to the fact that PsV production and purification are more effective than native virions.
