The energy unit of the cell is frequently referred to as adenosine triphosphate (ATP). It has proven challenging to assess its concentration in living mammalian cells in a non-invasive, real-time, and quantitative manner. Researchers created the fluorescent ATP indicator protein QUEEN-37C, which is ideal for determining the amount of ATP present in active mammalian cells. The luciferase assay was used to confirm the ratiometric fluorescence imaging's accuracy in estimating the absolute value of the ATP content. Since QUEEN-37C makes it possible to detect ATP concentration in a single cell, researchers were able to determine both its mean and its distribution among the population of cells, which showed that ATP concentration is tightly regulated in the majority of cells.
