The functioning of the brain is significantly influenced by neuromodulatory systems. Measuring spatiotemporally accurate neuromodulator release is necessary to comprehend how these systems alter the operating mode of target circuits. To enable optical recording of dopamine dynamics with high spatiotemporal resolution in behaving mice, researchers created dLight1, an intensity-based genetically encoded dopamine indicator. By simultaneously studying dopamine dynamics with pharmacological modification, electrophysiological or optogenetic stimulation, and calcium imaging of local neuronal activity, they showed the effectiveness of dLight1. Chronic monitoring of learning-related alterations in the striatal striatal millisecond dopamine transients was made possible by dLight1. They also employed dLight1 to image dopamine transients in the cortex that are relevant to learning and motor control that are geographically different and functionally heterogeneous.
