Nitric oxide plays diverse roles in asthma, serving as both an inherent regulator of airway function and an instigator of inflammation. Fractional exhaled nitric oxide (FeNO) stands as a dependable, quantitative, non-intrusive, uncomplicated, and secure biomarker for appraising airway inflammation in asthma. An investigation was conducted, comprising 264 asthma cases, discovered among the general adult population in Verona, Italy, between 2008 and 2010. This study targeted the identification of single nucleotide polymorphisms (SNPs) within candidate genes or gene regions associated with FeNO in asthma. Machine learning via the gradient boosting machine (GBM) technique identified 15 significant SNPs, among which SNP rs987314 within the family with sequence similarity 13 member A (FAM13A) and SNP rs3218258 within the interleukin 2 receptor subunit beta (IL2RB) gene regions displayed notable associations with FeNO in adult asthmatic subjects. These genes exert influence on various mechanisms affecting smooth muscle constriction, endothelial barrier function responses (FAM13A), and immune response processes (IL2RB), expanding the understanding of FeNO's role in asthma.
