    Product Information

    Product Overview
Membrane vesicles (extracellular vesicles; EVs) are nano-sized lipid bilayer vesicles released by cells. This product is membrane vesicles (extracellular vesicles; EVs) from Placental Amnion-derived Mesenchymal Stromal Cells (MSCs). It can be used in Drug Delivery Research; Regenerative Medicine Research and Wound Healing; Inflammatory Disease; COVID-19.
    Membrane Vesicle Introduction
Extracellular vesicles (EVs) are nano-sized lipid bilayer vesicles secreted by cells into the extracellular space. EVs play an critical role in cellular communication by transporting proteins, lipids as well as nucleic acids. They are present in biological fluids and are involved in multiple physiological and pathological processes. EVs have diverse biological activities, ranging from roles in development and homeostasis to cancer progression, which has spurred the development of EVs as disease biomarkers and drug nanovehicles.
    Origin Cell Name
Placental Amnion-derived Mesenchymal Stromal Cells (MSCs)
    Organism
    Human
    Cell Grown Condition
    Hollow fiber bioreactor
    Biosafety Level
    BSL 1
    Cell Viral Testing
The cells have been tested negative for HIV, HepA, HepB, HepC and Parvo-B-19.
    Cell Characterization
The cells express typical marker proteins of mesenchymal stem cells, and can be differentiated towards adipocytes, chondrocytes and osteoblasts.
    EV Enrichment Method
    Tangential flow filtration (TFF)
    EV Enrichment Procedure
EVs are enriched from cell culture supernatant using tangential flow filtration (TFF) with a modified Polyethersulfone (mPES) membrane with a MWCO of 300 kDa.

    Highlights
    Tissue Specific Delivery

    Product Quality Control

    Sterility Testing
    Negative
The EVs are bacteria and fungi free, which can be used for sensitive cell cultures.
    Mycoplasma Testing
    Negative
The EVs are mycoplasma free, which can be used for sensitive cell cultures.
    Endotoxin Level
    < 0.01 EU/mL (determined by LAL test)
    EV Marker
    CD81; CD9; CD63
    Anti-inflammatory Activity
    Positive
Treatment of mouse macrophage cells (RAW264.7) with lipopolysaccharide (LPS) induces the formation of nitric oxide (NO) formation indicating an inflammatory reaction. Addition of EVs derived from Bone Marrow-derived Mesenchymal Stromal Cells (MSCs) significantly reduces NO secretion, indicating positive anti-inflammatory activity of the EVs.
    Anti-fibrotic Activity
    Positive
Treatment of human fibroblasts (fHDF/TERT166) with Transforming Growth Factor beta (TGF-β1) induces the expression of alpha smooth muscle actin (α-SMA) indicating myofibroblast differentiation/activation, which is a key event in physiological and pathological tissue repair. Addition of EVs derived from Bone Marrow-derived Mesenchymal Stromal Cells (MSCs) significantly reduces α-SMA induction, indicating positive anti-fibrotic activity of the EVs.

    Product Handling and Storage

    Application
Wound healing studies; Anti-inflammatory therapeutics discovery and development; COVID-19 therapeutics discovery and development
    Handling Notes
Frozen EVs should be immediately transferred to -80°C upon receipt.
Note: Thaw the EVs on ice, centrifuge before opening the tube to ensure that the solution is collected at the bottom of the tube. Then, mix carefully by pipetting up and down and aliquot for further use. Avoid freeze/thaw cycles as it may result in irreversible damage to the EVs.
Disclaimer: We cannot guarantee the activity if the EVs are not handled according to procedure.
    Use Restrictions
The EVs products are for Research Use Only (RUO), and not for clinical use in humans for therapeutic purposes.
    Shipping
    Dry ice
    Storage
Store the EVs at -80°C (for up to 6 months) until use.
After thawing, store the EVs at 4°C for a maximum of 1 day.
