Workflow of Cancer Cell Panel Screening Service
- Human cancer cell lines were seeded into 384-well microplates. After being incubated for the entire night, the cells were treated with compounds and then re-incubated for the experiment at a later time.
- ATP is measured using CellTiter-Glo. It is commonly acknowledged that ATP is a reliable indicator of living cells. The ability of cells to manufacture ATP is lost when their membranes break, and endogenous ATPases quickly use up whatever ATP that is still in the cytoplasm. The ATP detection reagent is made up of luciferin as a substrate, detergent to lyse the cells, ATPase inhibitors to stabilize the ATP released from the lysed cells, and stable form of luciferase to catalyze the reaction that produces light photons.
- The studied datasets integrate high throughput drug sensitivity screening with molecular characterization of cancer cell lines. Cell viability at various escalating drug doses was assessed for each drug-cell pair examined in a dataset, and the results were compared to an untreated control to determine the percentage of viability. It is desirable to obtain a single number summarizing the sensitivity of a certain cell line to a drug treatment in order to learn drug response predictors.
