Numerous physiological functions, such as the sleep-wake cycle, learning and memory, cardiovascular health, and immunological responses, depend on extracellular adenosine (Ado). Through the activation of several G protein-coupled Ado receptors, Ado functions at the synaptic level in the brain as a neuromodulator or a homeostatic modulator. The genetically encoded GPCR-Activation-Based (GRAB) sensors were made to measure the dynamics of a number of neuromodulators, including acetylcholine, dopamine, and norepinephrine, and they have been used in a variety of in vivo settings. The GRAB-Ado sensor is a genetically encoded device. In living mice, GRAB-Ado can quickly and with hitherto unheard-of sensitivity, stability, and temporal resolution report endogenous Ado dynamics after seizures and hypoxia.
