Serotonergic transmission in the brain is essential for a variety of physiological functions, including mood regulation, reward processing, and the homeostatic management of sleep-wake cycles. In fact, almost all mental disorders have been treated with medications that target central serotonergic activity, with the finest example being the use of selective serotonin reuptake inhibitors (SSRIs) for depression. Despite this, and in part because we are unable to quantify 5-HT in vivo with high sensitivity and spatiotemporal precision, our understanding of cell-specific 5-HT signaling during activities is severely deficient. Researchers created a genetically encoded GPCR-Activation-Based 5-HT (GRAB5-HT) sensor with excellent sensitivity, selectivity, subsecond kinetics, and subcellular precision to directly visualize the release of 5-HT. With its ability to detect 5-HT release in a variety of physiological and pathological circumstances in both flies and mice, GRAB5-HT offers fresh perspectives on the dynamics and workings of 5-HT signaling.
